pGLO Non-Fluorescent Mutant

We generated this in our lab by mistake. The Bio 6B Special Projects team was using site-directed mutagenesis to alter the GFP coding region, but by a slight miscalculation, we ended up with a mutant that does not produce a fluorescent protein at all:

We're not using this plasmid, but we are using a related plasmid, pARO180.  The sequence of pARO180 is not available, so this is as close as we can get. Unfortunately, there are some differences between these two related plasmids in terms of the orientation of the DNA fragments used to create the plasmid, so the restriction map is different between the two.

Notes about formats

Data formats

Sequence data can be used in a variety of formats. The simplest is plain text (for example, a plain text version of pGLO), but a continuous stream of letters can be hard to work with. It's also common to show sequence data with a space every 10 letters, and to add numbers for easier navigation. Sequence analysis software will generally ignore the spaces and numbers.

In most cases, you can include a name or other information at the beginning of a sequence. For example:

EMBOSS Seqret from EMBL-EBI. Enter a sequence in one format, get it out in another. For example, if you need to eliminate the numbers, spaces, and line breaks, this tool will do that.

Benchling. I use this for all kinds of sequence analysis. You need to make a free account to use it, but it's worth the effort.
